Mark: I don't really have a family. Derek was my family

Mark: Derek's damaged goods. I damaged him.

Derek: He was like my brother. I have four sisters. Four very…annoying sisters. Mark was my brother, it’s hard.

Mark: I made an excellent best man.
Derek: (turns to Burke) Worst case scenario, I sleep with your wife in 10 years.
Episode 3.24 - Testing - 1, 2, 3

Mark: I'm the one that could use a friend. I said that you could use a friend. I could use a friend.
Derek: Look. Mark---
Mark: I know. I didn't come to Seattle for Addison. I didn't come to Seattle to be chief. I came to Seattle for you. Okay? I came to Seattle to get you back.
(There's an awkward pause. Mark looks embarrased and Derek looks like he might start laughing.)
Mark: I know. I wanna take it back now, but I already said it so... (chuckles)
Derek: (chuckles) Yeah. Maybe we should have a drink.
Mark: We could.
Derek: Yeah. Not today.

Derek: You're like the worst, most juvenile human being I've ever met in my life.
Mark: You know the first step to recovery is admitting you have a problem?
Derek: How many nurses have you slept with this week?

Mark: Hey, if you were sleeping with Rose, I could understand why you're spending time with her, but you're not. And it's... it's just I thought that uh... you know. I thought it was just gonna be you and me!
Derek: Having sex?
Mark: After! After Addison, after Meredith. I have been patient. I have waited when you partnered up with these chicks, but now they're over. And it's supposed to be just us! You and me; two guys on the prowl, on the hunt.
Derek: You're having trouble getting laid. And you need my help.

Mark: I can get laid. I can get laid whenever I want. I DO get laid whenever I want. That's the point. Women are everywhere... I only have one person I can talk to.
Derek: That's sweet.
Mark: Shut up!

Mark: One woman 10 points, two women same time 20 points. You get out in the morning before anybody wakes up; 5 point bonus.
Derek: You are serious.
Mark: You need a little spice in your life, man. I'm just trying to help.
Derek: I am truly a worse human being for knowing you.

Mark: You know what, I'm done. I give up. If I hadn't fallen for little Grey, you would have found some other way not to respect me, you always have. To you, I'm always gonna be the charity case the Shepherds had to take in. I got the bleeder.

Mark: You know that's the first actual compliment you've ever given me?
Derek: It wasn't a compliment, it was a simple statement of fact.
Mark: See, why do you do that? Why can't you just give a person a compliment?
Derek: If you have to ask for a compliment, it's not a compliment. Are you asking for a compliment?
Mark: Yes!
Derek: Then it's not a compliment!
5.20 - Sweet Surrender

Mark: I'm thinking about buying a house.
Derek: Good for you. Meredith and I are gonna go to city hall this afternoon. (smiles) We're getting married.
Mark: Really?
Derek: Mmmm.
Mark: That's great news.
Derek: I know, and if I could just save Stevens...
Mark: And you're allowed to be happy. You're allowed to get married. You've earned that right. Congratulations you lucky bastard. Come here. Give me a hug. (they hug)
Episode 5.24 - Now or Never

#2 ~ Onscreen or off, these two guys were meant to be friends!
